The paternal grandparents of Mary Gregory Litsey were Richard and Agnes West Gregory of King William County, Virginia. They owned the Mount Vernon estate which they later deeded to Augustine Washington, the father of George Washington. Richard and Agnes's son, also named Richard, married Anne Vaughn and moved to Kentucky from Mecklenburg County, Virginia in the late 1780s. They were one of the pioneer families that settled in the Mooresville area of Washington County, Kentucky. They were also among the early members of Pleasant Grove Presbyterian Church. Their home was noted for its purity, culture, and social standing.

Richard and Anne's daughter, Mary Gregory, married Randal Litsey, son of Anthony and Susannah Slack Litsey, on December 30, 1797.

Mary was the girlhood friend of Nancy Hanks, the mother of Abraham Lincoln. When Nancy Hanks and Thomas Lincoln were married, Mary helped make Nancy's wedding dress. Mary and her husband, Randal Litsey, attended the wedding of Thomas Lincoln and Nancy Hanks on June 12, 1806 at the home of Nancy's cousin, Francis Berry, in Washington County, Kentucky.
